Refractometer Calibration Standards

Rudolph Research Analytical works with Cargille Laboratories to provide a range of refractometer calibration standards.

All Calibration standards are:

  • Supplied in 28ml bottles
  • Supplied with a calibration certificate traceable to the National Institute of Standards and Technology (NIST)
  • Shelf life of unopened bottle is 5years
  • Shelf life of opened bottle is 2.5 years
  • Fluids calibrate at temperatures between 20°C and 40°C are accurate to 0.0001 Refractive index

Choosing a Refractometer Calibration Fluid

Rudolph Research Analytical offers two types of calibration fluids:

  • NIST traceable refractive index fluids
  • Brix standards made from sugar and water

Technical Considerations

Description of Fluids Refractive index fluids are made from chemical compounds such as methylene iodide, silicon oil and alpha bromonapthalene. These chemicals are chosen as they are non toxic though some of the fluids do have an unpleasant odor (alpha bromonapthalene = mothballs). Brix fluids are a mixture of sucrose and water. Brix fluids
They are so safe you could drink them. They are just sugar and water.
Traceability of Fluids Refractive index fluids are provided with a certificate showing traceability to a national standards body - NIST. Brix fluids are made using the method specified in ICUMSA and are provided with a non-traceable certificate of analysis. Refractive Index fluids

Shelf Life    Refractive index fluids have a rated life of 5 years unopened, 2.5 years opened. Care should be taken that they are not frozen, but otherwise they can be stored at room temperature

Temperature Dependency All fluids are temperature dependant. The refractive index fluids have a different temperature dependency than the temperature compensation setting on a refractometer and so can only be used on a temperature CONTROLLED refractometer. Application Considerations

   Refractive Index Fluids  Brix Fluids     Advantages
Type of refractometer Temperature controlled Refractometer is required. If the refractometer does not have a method of controlling temperature then the refractive index fluids will not work. This applies to all hand refractometers and also Abbe refractometers that do not have a water bath attached. It also applies to a few cheaper bench refractometers. For these refractometers the only choice is the Brix fluids. Brix fluids
They can be used to calibrate hand refractometers as well as bench models.
 Ease of Use  The refractive index fluids are supplied in a glass bottle with a glass dropper stick. The fluids last a long time (2.5 years minimum) but care must be taken to ensure the bottle is not contaminated. In general we would recommend that use of these fluids is restricted to users with some laboratory training. The Brix fluids are supplied in a dropper bottle and are more regularly replaced. This means they are much simpler to use for non-lab trained people. Brix fluids

Temperature Range  If the laboratory procedures specify measurement be controlled to a temperature other than 20°C then refractive index fluids calibrated at that temperature must be used.  Brix is, by definition, calculated at 20°C.  Refractive Index fluids

Very General Shortcut- Use Like for Like

Use a fluid that is like the sample to be measured

  • If the results are reported in Brix then use Brix standards
  • If the results are reported in Refractive Index then use Refractive Index standards.

J57TPN AUTOMATIC REFRACTOMETER

The J57TPN is intended for use in hospital pharmacies to monitor the quality of parenteral nutrition mixtures and ensure compliance with USP<797>. It is also useful in preventing the diversion of narcotics and other controlled materials. The J57TPN provides a fast, easy to use way to check on materials returned from the operation theatre or incorrectly issued from the pharmacy.


Application

The first decade of the 21st Century has seen dramatic changes in health system pharmacies. New regulations such as USP<797> are forcing compounding pharmacies to do quality checks that were previously only required of pharmaceutical manufacturers. The J57TPN from Rudolph Research Analytical is designed to comply with this new regulatory environment.


Drug Diversion Monitoring

The J57TPN also doubles as a refractometer to monitor potential drug diversion issues. Ever more strict laws mean a health system must be aware of what is happening with controlled substances. Measuring operating theater returns and similar movements of controlled substances can be a key part of an organization's policy on meeting these laws.


Automatic Measuring System

The J57TPN features a fully automatic measuring system. The operator simply places the sample on the prism, presses a button and the result is displayed on a 7.5cm x 10cm backlit LCD. There are no shadow lines to match and there is no eyepiece requiring operator determination or manual adjustment.


Pharmacy Relevant Calculations

The J57TPN has built-in comparison and calculations. The user enters the % Dextrose and % Amino Acid and the instrument displays both the calculated and measured result, making it easy for an operator to see if the material is within specification.


Electronic Temperature Control

Refractive Index is a temperature sensitive property. For many years pharmacies largely ignored this fact or used sugar based temperature correction. Pharmaceutical manufacturers who were constrained by USP guidelines were not able to ignore this as USP<831> clearly states that they had to measure at a controlled temperature, generally 20°C. Now that pharmacies are under similar pressure, temperature sensitivity also must be considered. Fortunately with the J57TPN, old style circulating baths are a thing of the past and sucrose based mathematical approximations are not needed because of the instrument's built in electronic heating and cooling which ensures the sample is measured at 20°C every time.


Electronic Record Keeping Option

In the modern pharmacy, making a test is not enough. The pharmacy has to prove not only that they did the test, but who did it, when they did it, and if the instrument was correctly calibrated. The J57TPN MMC helps with this by providing full electronic records of every measurement, in line with US FDA Regulation 21 CFR Part 11. As well as making a system much easier to use, the MMC option can also assist by enabling a pharmacy to avoid locating printers in a clean room, something that is always difficult to manage under USP<797> guidelines.

Overview

This is a system just like J257 but without the top cover. Model J257 has a range between 1.26 to 1.72RI and 0 - 100Brix. The accuracy is +/- 0.0001RI / +/-0.1Brix. The resolution is 0.0001RI and 0.01Brix. The temperature control of the sample can be done in either 20 deg C or 25 deg C. This would satisfy both USP and EP needs. The communication interface is through 2 RS232, one parallel printer port and one auxiliary port.

The same system can also be configured for the reading of Butyro scale. The Butyro has to be analysed in 40 deg C and the J57WR can be set for 20 deg C & 40 deg C. This would allow this system to be used in any food company where the analysis would either be for Sucrose or for Butyro.

When measuring sucrose users have the option of correcting for temperature variation but that option does not work for most chemical companies. Users not measuring sucrose or users looking for better accuracy than provided by temperature correction had to spend the extra money for a circulating water bath and then clean and maintain it. The J57WR’s built in electronic temperature control provides the best of both worlds, the absolute accuracy, regardless of sample type, of a temperature controlled instrument with the convenience of operating without a water bath.


Wide Measuring Range

The J57WR was designed for chemical measurement, not food. This means a very wide measurement range from 1.26 - 1.70 enabling the instrument to measure materials like fluorocarbons and aromatic chemicals that are beyond the range of most refractometers.


Easy to Clean Measurement Surface

Regardless of the specified instrument accuracy the real world performance will always depend on how well the instrument is cleaned between samples. The J57WR addresses this issue by providing a very flat easy to clean measurement surface with no corners or crevices that tend to trap samples in instruments designed for laboratory use.

High Durability Sapphire Prism

When a traditional refractometer is used in a high throughput application, prism replacement becomes a regular task. Traditional Abbe refractometers have glass prisms which are much softer than the sapphire prism of the J Series and thus more prone to scratching and general wear and tear.


Lowest Whole of Life Cost

Users in high throughput chemical applications have traditionally used Abbe refractometers due to their low purchase price. Changes in relative prices between refractometers and optics and the elimination of a water bath mean the J57WR now has a much lower whole of life cost than an Abbe refractometer.


Calibration

The J57WR Automatic Refractometer offers 1 or 2 point calibration. The instrument stores the date and the time of the last calibration in line with ISO and similar standards. Regardless of how many times the refractometer is calibrated it can always be restored to the default factory calibration.

The J Series customizable display allows the instrument to be configured to exactly what is required for each application. The customizable keypad can be "locked out" or just a few keys can be operable, i.e. the measure key, print key, zero key, customer specified sample keys or any combination thereof. The lockout function, calibration and other functions are all password protected.

J57 HA Refractometer

Electronic Temperature Control

Brix readings are very sensitive to temperature changes. However these changes in BRIX readings can be minimized by using the ICUMSA temperature correction for sucrose, but only minimized, not eliminated. The ICUMSA Temperature Correction Table was developed for a sucrose and water solution and the further from pure sucrose and water the sample is, the more inaccurate Temperature Correction becomes. Temperature Correction also requires that the sample temperature remain stable during the measurement. So a room temperature sugar solution measurement works reasonably well, but if a sauce sample comes from a kettle or a refrigerator, temperature correction will be almost useless. The solution to measuring hot samples has always been to control the sample temperature with a circulating water bath and then clean and maintain the water bath. The J57HA’s built in electronic temperature control provides the best solution for these real world problems: absolute accuracy regardless of sample type and excellent thermo stability no matter what the sample or environmental temperatures are.
